We discovered that osteoarthritic synovial fluid microenvironment instigates site-specific changes in the rRNA 2β-O-me profile of primary human articular chondrocytes.

The 2β-O-me status of 5.8S-U14 (one of the identified differential 2β-O-me sites) was targeted by depleting the level of its guide snoRNA (SNORD71). This resulted in an altered ribosome translation modus and promoted translation of COL1A1 mRNA.
These data provide a novel concept of how articular chondrocytes employ rRNA 2β-O-me heterogeneity as an epitranscriptomic regulatory mechanism to control translation of osteoarthritis-relevant mRNAs.

In this study, subregional 3D JSW measured on weight-bearing CT was significantly more responsive to temporal changes than 2D JSWx on radiographs in knees with or at risk for knee osteoarthritis. Use of 3D JSW on WBCT could enable improved detection of OA structural progression.
